AllGPCRs,includingGPR68,aresinglepeptidespossessingseventransmembranedomainswithanextracellularN-terminusandanintracellularC-terminus1.GPR68isexpressedinseveraltissuesincludingthespleen,testis,smallbowel,brain,heart,lungandkidneys.Itsharesstronghomology(49-54%)withthehumanreceptor GPR42.
GPR68isprotonsensitiveinthepHrangeof6.8-7.8andactsasaprotonactivatedreceptor.GPR68stimulatesinositoltriphosphate(IP3)formationtherebyreleasingintracellularcalciumandactivatingthemitogenactivatedproteinkinase/extracellularsignal-regulated(ERK)kinasecascade.Inaddition,GPR68receptorregulatesNHE3andH+-ATPase,inresponsetoextracellularpHchangesinculturedHEK293renalcells.Thisactivationrequiresaclusterofhistidineresiduesintheextracellulardomainsofthereceptorandprotein-kinaseC(PKC)activation4.
